Edman degradation

Edman degradation is a method of sequencing peptides by sequentially removing and identifying one amino acid at a time from the N-terminus using phenyl isothiocyanate. Developed by Pehr Edman, it requires a free N-terminus and is practical for up to about 30 residues, with an upper limit of roughly 50 to 60. Mass spectrometry has largely supplanted it.
