PEGylation explained: attaching polyethylene glycol to a peptide

Short answer

PEGylation is the covalent attachment of one or more polyethylene glycol (PEG) chains to a peptide or protein. PEG is a flexible, water-soluble polymer of repeating -CH2-CH2-O- units. Attaching it greatly increases the size of the molecule in solution, and it can shield the peptide from proteases and from recognition by other proteins. Reviews by Harris and Chess (2003) and by Veronese and Pasut (2005) describe how the approach developed and the chemistry behind it.

How PEG is attached

PEG is supplied as activated derivatives that react with particular groups on the peptide. Common targets are primary amines, at the N-terminus or on lysine side chains, and free cysteine thiols, which react with maleimide-activated PEG. Site-specific attachment, at one defined residue, gives a more uniform product than random coupling to every available amine.

PEG reagents vary in length, usually described by average molecular weight, and in shape: linear or branched.

Why PEG makes the molecular weight an average

Synthetic PEG is a polymer with a distribution of chain lengths, so a PEGylated peptide is a mixture of closely related species rather than one molecule with one exact mass. Its mass spectrum shows a series of peaks about 44 Da apart, one ethylene-oxide unit each, instead of a single peak. Databases therefore often have no single formula or molecular weight for such a product. This is why the Sable /ref page for PEG-MGF shows formula and molecular weight as not on file rather than quoting one figure.

What changes in the lab

Size and solubility. The PEG chain binds a lot of water, so the conjugate behaves as a much larger molecule than its mass alone suggests. That affects size-exclusion chromatography and filtration.

Chromatography. PEGylated peptides give broader HPLC peaks than the parent peptide because of chain-length heterogeneity. Purity is often assessed by several complementary methods.

Mass spectrometry. Identity checks look at the peptide part and the average PEG mass, rather than a single monoisotopic peak.

Activity in assays. A PEG chain near a binding site can reduce how well the peptide binds its partner. This is one reason the attachment site matters.

PEGylation compared with other half-life strategies

PEGylation is one of several structural strategies studied for extending how long a peptide persists. Others include attaching a fatty-acid chain, which promotes reversible binding to albumin (see the lipidation explainer), and attaching an albumin-reactive linker. CJC-1295's DAC is a maleimide-containing linker designed to bind albumin covalently. These are chemically distinct approaches and should not be confused when you read a structure.

At Sable

PEG-MGF is listed with its structure description and literature sources. Because of PEG heterogeneity, the certificate of analysis for a specific batch, available for every lot at /coa, is the authoritative record of what was measured. Sable Peptides has been operating since 2019 and supplies every compound mentioned here as laboratory research material only. Nothing on this page is advice on use in people or animals, and no administration or medical-use information is provided.

Key terms

Polyethylene glycol (PEG)
A water-soluble polymer of repeating ethylene-oxide units.
Polydispersity
A spread of chain lengths, and therefore masses, within one polymer sample.
Maleimide
A reactive group that couples selectively with free thiols, such as cysteine side chains.
Hydrodynamic size
How large a molecule behaves in solution, including bound water. For PEG conjugates it is much larger than mass alone suggests.
Site-specific conjugation
Attaching PEG at one defined residue, rather than at random.

Common questions

Why is there no single molecular weight for a PEGylated peptide?

PEG is a polymer with a range of chain lengths, so the product is a distribution of masses about 44 Da apart.

Is PEGylation the same as lipidation?

No. PEGylation attaches a polyethylene glycol polymer; lipidation attaches a fatty-acid chain.

Which Sable compound is PEGylated?

PEG-MGF.
