HPLC vs Mass Spectrometry (LC-MS) for Peptide Testing

HPLC vs Mass Spectrometry (LC-MS) for Peptide Testing is supplied for laboratory research only. Key facts are below; stored as lyophilised powder.

A CoA for every lot; each vial carries a printed lot number that matches it. Ask with the lot number and we send it.

The short answer

HPLC measures purity and mass spectrometry confirms identity. LC-MS combines them, putting a mass to each chromatographic peak, which is the strongest routine check on a research peptide.

How the MS part works

Electrospray ionisation turns large, polar molecules such as peptides into multiply charged gas-phase ions (Fenn et al., 1989). The analyser measures m/z, and software reconstructs the neutral mass. High-resolution instruments resolve isotopes and can spot +1 Da (deamidation) or +16 Da (oxidation) changes (Aebersold & Mann, 2003).

How the HPLC part works

Reversed-phase separation with UV detection at 214–220 nm reports the main peak's share of the total area (Mant et al., 2007). LC-MS often uses formic acid instead of TFA, because TFA suppresses ionisation.

What a certificate should show

At minimum, the HPLC purity with its method, plus the observed and expected mass. LC-MS data, when supplied, is the stronger evidence.

How much does HPLC vs Mass Spectrometry (LC-MS) for Peptide Testing cost in the UK?

Frequently asked questions

What does HPLC tell you?

How much of the UV-absorbing material is the main species: purity.

What does mass spectrometry tell you?

Whether the molecular mass matches the expected sequence: identity.

What is LC-MS?

Liquid chromatography coupled directly to a mass spectrometer, so each HPLC peak can be assigned a mass.

Why do MS spectra show several peaks for one peptide?

Electrospray ionisation produces several charge states (M+2H, M+3H and so on), which are deconvoluted to one mass.

Which test matters more?

Both. A pure wrong peptide and an impure right peptide each fail one of the two tests.
