Kisspeptin-10, gonadorelin and triptorelin: how do these reproductive-axis peptides differ?

Kisspeptin-10, gonadorelin and triptorelin act at different points on the reproductive hormone axis. Kisspeptin-10 is the C-terminal decapeptide of kisspeptin and acts at the KISS1 receptor (GPR54) on GnRH neurons. Gonadorelin is synthetic GnRH. Triptorelin is GnRH with a D-tryptophan in position 6.

Where does each peptide act?

  • Kisspeptin-10: ligand of KISS1R (GPR54), upstream of GnRH release. Loss-of-function GPR54 mutations were linked to failure of puberty in 2003.
  • Gonadorelin: the native GnRH decapeptide, a ligand of the GnRH receptor on pituitary gonadotrophs.
  • Triptorelin: a GnRH-receptor agonist analogue. The D-Trp6 substitution stabilises a β-turn and slows enzymatic cleavage at the Gly6 position.

How do their sequences compare?

Gonadorelin and triptorelin differ at one position only, Gly6 → D-Trp6. Kisspeptin-10 is unrelated in sequence and belongs to the RF-amide family: its C-terminus is Arg-Phe-NH2.

What laboratory work has used them?

Kisspeptin-10 has been applied to GnRH neurons and to pituitary cells from livestock species in vitro. Gonadorelin products have been compared in veterinary reproductive physiology, and chromatographic methods for telling GnRH derivatives apart have been published.

Frequently asked questions

What is the difference between gonadorelin and triptorelin?

Gonadorelin is native GnRH. Triptorelin replaces Gly6 with D-Trp, giving a more stable GnRH-receptor agonist analogue.

Is kisspeptin-10 a GnRH analogue?

No. It is the C-terminal 10 residues of kisspeptin and acts at KISS1R (GPR54), upstream of GnRH neurons.

What does pGlu mean in the gonadorelin sequence?

Pyroglutamate, a cyclised N-terminal glutamine or glutamate that blocks the free amine.
