## Net peptide content vs gross weight

*Last updated: 2026-10-10 · Research use only. Not for human or veterinary use.*

## Short answer

Gross weight is the mass of lyophilised powder in a vial. Net peptide content is the percentage of that powder that is the peptide itself, after counter-ions (such as trifluoroacetate or acetate) and bound water are accounted for. A typical literature range for synthetic peptides is 60–90% \[1\]; this is not a Sable specification. It is independent of HPLC purity. \[1\]

## At a glance

| Term | Definition / specification |
| --- | --- |
| Gross weight | Total mass of lyophilised solid |
| Net peptide content | Peptide mass ÷ gross mass × 100 |
| Main non-peptide components | Counter-ions, residual water, trace solvents |
| Typical literature range | 60–90% net peptide content [1]; not a Sable specification |
| Typical methods | Amino acid analysis, elemental nitrogen analysis, quantitative NMR |
| Relation to purity | Separate measure; a high-purity lot can have modest net content |

## Why the two numbers differ

Basic residues (Lys, Arg, His, the N-terminus) carry positive charges that are balanced by anions picked up during cleavage and purification. Lyophilised solids also retain some water. Both add mass without adding peptide.

## Reading them together

Multiplying purity by net content gives an approximate fraction of the target sequence in the powder. Reference-standard guidance handles the two as distinct attributes.

## FAQ

**Is net peptide content the same as purity?**

No. Purity compares peptide species with each other; net content compares peptide with everything else in the vial.

**Which method measures net content?**

Amino acid analysis after acid hydrolysis is the classic method; nitrogen analysis and quantitative NMR are also used.
