# Container closure integrity: how a sealed vial keeps a peptide dry

Container closure integrity (CCI) is the ability of a vial, stopper and crimp seal to keep moisture, oxygen and microbes out. For a lyophilised peptide, a leak mostly means moisture getting in, which speeds degradation. Labs test CCI by dye ingress, vacuum decay, helium leak or headspace gas analysis.

## Which test methods are used?

| Method | Principle | Type |
| --- | --- | --- |
| Dye ingress | Vial immersed in dye under vacuum/pressure; dye entry seen or measured | Probabilistic |
| Microbial ingress | Vial challenged with bacteria; growth inside indicates a leak | Probabilistic |
| Vacuum decay | Pressure rise in a test chamber reveals leaks | Deterministic |
| Helium leak | Helium tracer measured by mass spectrometer | Deterministic, most sensitive |
| Headspace analysis | Laser measures oxygen, CO2 or moisture inside the vial | Deterministic, non-destructive |

## Why is CCI a whole-package question?

Integrity depends on vial dimensions, stopper material and fit, and crimp force together. A holistic approach to package design, and recent industry guidance for products that are hard to test, both treat CCI as a lifecycle property, not a single test.

## What can a lab check on receipt?

- The crimp is intact and cannot be rotated easily.
- The stopper is seated flat, not raised or tilted.
- The cake or powder is dry and not collapsed or stuck to the stopper.
- No cracks or chips at the vial neck.

## Does a vacuum in the vial matter?

Many lyophilised vials are stoppered under partial vacuum or nitrogen. A slight hiss when piercing is normal; its absence does not on its own prove a leak, but together with a wet or collapsed cake it is a warning sign. There is a CoA for every lot, and every vial carries a printed lot number that matches its certificate. A CoA is emailed within 2 hours of a request.

## Frequently asked questions

### What is container closure integrity?

The ability of a vial, stopper and seal to prevent ingress of moisture, gases and microbes.

### Which CCI test is most sensitive?

Helium leak detection is generally the most sensitive deterministic method.

### What visual signs suggest a leaking vial?

A loose crimp, raised stopper, cracks at the neck, or a wet or collapsed cake.
