# UV detector

A UV detector measures the absorbance of ultraviolet light by compounds eluting from an HPLC column. Peptide bonds absorb strongly near 214 nm, and aromatic residues such as tryptophan and tyrosine absorb around 280 nm. Detection at 214 to 220 nm is used for peptide purity because all peptides respond.
