# Reversed-phase chromatography

Reversed-phase chromatography uses a hydrophobic stationary phase, such as C18-bonded silica, and a polar mobile phase, usually water and acetonitrile with an acid modifier. More hydrophobic molecules are retained longer. It is the dominant mode for analytical and preparative separation of peptides because of its high resolving power.
