# Orthogonal protection

Orthogonal protection is a strategy in which different protecting groups in the same molecule can be removed independently using distinct conditions, such as base, acid, or palladium catalysis. In Fmoc/tBu peptide synthesis, the base-labile Fmoc group is removed each cycle while acid-labile side-chain groups remain intact until final cleavage, enabling controlled chain assembly and selective modification.
