Glossary

# N-methylation

N-methylation is the replacement of a backbone amide hydrogen with a methyl group. In peptides it removes a hydrogen-bond donor, can increase membrane permeability and resistance to proteases, and restricts conformation. N-methylated residues are harder to couple in synthesis and often require strong coupling reagents such as HATU.
