# Kaiser test

The Kaiser test is a colorimetric test based on ninhydrin used to detect free primary amines on resin during solid-phase peptide synthesis. A blue colour indicates unreacted amines, meaning coupling is incomplete, while a colourless or yellow result suggests complete coupling. It is unreliable for secondary amines such as proline.
